Genoma de homem já extinto é sequenciado em detalhes

Cientistas da Universidade de Copenhague, Dinamarca, são os primeiros a reconstruir o genoma de um ser humano extinto em detalhes. A técnica inovadora poderá ser usada em vestígios encontrados na natureza ou guardados em museus para recuperar fenótipos humanos de culturas extintas – explicando o fluxo de populações pré-históricas pelo mundo. Além disso, permitirá uma compreensão maior sobre a hereditariedade e o risco de doenças associadas aos nossos antepassados.

Eske Willerslev, que liderou a pesquisa, e sua equipe já tinham chamado atenção no ano passado quando anunciaram a reconstrução completa do genoma mitocondrial de um mamute e um homem pré-histórico. No entanto, a pesquisa atual é inédita porque conseguiu reconstruir 80% do genoma nuclear através de restos fósseis. A partir das sequências do genoma, a equipe está conseguindo reconstruir a imagem de um indivíduo do sexo masculino que viveu na Groenlândia há 4 mil anos.

A descoberta foi obtida a partir das análises de um tufo de cabelo que pertenceu a um homem – apelidado de Inut – da cultura Saqqaq, ao noroeste da Groenlândia, e que estaria no Museu Nacional de História Nacional da Dinamarca desde que foi achado em escavações na década de 1980. “Depois de conseguir permissão do museu, nós analisamos o DNA do cabelo usando diversas técnicas, descobrindo que se tratava de cabelo humano. Durante muitos meses, estivemos em dúvida se nossos esforços não seriam em vão. Entretanto, por meio do trabalho duro de uma equipe de pesquisadores internacionais, finalmente conseguimos fazer o seqüenciamento genético de um homem extinto”, disse Willerslev.

A reconstrução dá uma boa ideia de como o homem pré-histórico da Groenlândia seria fisicamente – inclusive sua tendência à calvície, olhos castanhos, pele morena, sangue tipo A+, dentes da frente arqueados, quanto era geneticamente adaptado às baixas temperaturas e de que forma estava predisposto a certas doenças. A pesquisa também revela que esses ancestrais chegaram à América pelo norte da Sibéria entre 4400 e 6400 anos atrás, em uma onda de migração independente das dos americanos nativos.

“Os trabalhos anteriores para reconstruir o genoma do mamute resultaram em uma sequência de falhas devido a danos no DNA, porque a tecnologia ainda era recente. O genoma do Inuk é comparável em qualidade ao de um homem moderno”, diz Willerslev.

O pesquisador afirma que o achado pode ser uma ajuda significativa para arqueólogos e outros pesquisadores que tentam decifrar o que aconteceu com culturas já extintas. Para fazer isso, basta material orgânico – ossos ou cabelos guardados em peças de museus ou encontrados em sítios arqueológicos. Anteriormente, o DNA precisava estar congelado.
